The manual dissociation procedure described in the EvercodeTM FFPE Nuclei Preparation User Guide is the preferred method for FFPE nuclei preparation. The workflow is also compatible with the Miltenyi® Biotec FFPE Tissue Dissociation Kit for RNA Profiling (PN: 130-134-089). If using the Miltenyi kit, follow the Miltenyi’s protocol as written and stop at Section 2.2, Step 14. Then, continue with the Evercode WT FFPE Nuclei Preparation User Guide starting at Section 2, Step 15. Dissociated nuclei must be stored in Parse’s Nuclei Storage Master Mix to be compatible with downstream Evercode WT FFPE kits.
