We recommend a scroll input thickness of 25-50 µm. In addition, for highest data quality, we recommend first shaving off the outer edge of the FFPE block that has been exposed to air and then cutting the scroll(s) to be used for input into the nuclei dissociation process. For FFPE blocks that are a few years old or older, we recommend ideally shaving off approximately 25-50 µm from the exposed outer surface prior to collecting material for the assay.
Sections thinner than 25 µm can still produce usable data; however, cutting below this threshold may increase nuclear membrane damage, which can require additional scrolls to recover enough intact, dissociated nuclei.
