The CRISPR Detect kit is an add-on to the Evercode Whole Transcriptome kit. During cDNA amplification, barcoded transcripts are amplified, and CROP-seq hU6-sgRNA-polyA transcripts are enriched with primers specific to the human U6 promoter (hU6). Each sublibrary is then split to create Whole Transcriptome and CRISPR sequencing libraries, which can be sequenced together. After sequencing, the Parse Biosciences Analysis Pipeline assigns reads that share the same barcode combination to a single cell and associates CRISPR guides to the appropriate cell.
