The following equipment is required to run a Parse Biosciences EvercodeTM kit.
- Parse Biosciences Magnetic Rack (SB1004)
- Magnetic rack for 1.5 mL tubes.
- Magnetic rack for 15 mL conical tubes1: Permagen® (MSR6X15) or Millipore Sigma® (LSKMAGS15) or Stemcell Technologies® (18103)
- Ring Plate Magnet2: Permagen (s500) or Alpaqua® (A000405)
- Bar Magnet3: Permagen (MSPU650)
- Centrifuge with Swinging Bucket Rotors with adaptors for 1.5 mL tubes4, 15 mL tubes and 96 well plates and capable of reaching 4°C.
- Microcentrifuge compatible with 1.5 mL tubes and 0.2 mL strip tubes.
- Thermal Cycler5 compatible with semi-skirted 96 well plates and 0.2 mL PCR tubes with up to 100 μL of sample volume, sample heating and cooling from 4-98°C, and a heated lid capable of 30-105°C.
- Water bath, thermomixer, heat block, or bead bath capable of holding temperature at 37°C.
- Two heat blocks6: with a range of temperature from 48°C to 68°C and compatible with 1.5 mL, 2 mL, and 5 mL tubes.
- Hemocytometer or other cell counting device7.
- PCR tube rack capable of holding semi-skirted 96 well PCR plates and a tight fitting lid.
- Plate seal applicator capable of adhering plate sealing films to 96 well plates.
- Pipettes: Single channel P20, P200, P1000 and 8 or 12-channel P20, P200.
- Vortex and adapter for 96 well plates that can be set to 800-1000 RPM.
- QubitTM Flex Fluorometer
- 2100 Bioanalyzer or 4200 TapeStation System
- Mr. FrostyTM Freezing Container or styrofoam cooler8
- Freezers: -20°C, -80°C and 4°C.

4 The 1.5mL tube adapter is needed when using Evercode Cell Fixation, 12rxn or Evercode Nuclei Fixation, 12rxn which utilizes 1.5mL tubes.

7 An automated cell counter compatible with fluorescent dyes (e.g., AO/PI) is required for Evercode Low Input v3, Evercode Low Input v4, and Evercode WT v4 workflows. For all other workflows, an automated cell counter or hemocytometer may be used. Prior to use with a new cell type, we recommend validating the automated cell counter by comparing to a manual count on a hemocytometer.
8 Fixed samples should be slowly cooled for storage at -80°C. A Mr. Frosty Freezing Container should be used when fixed samples are stored in 1.5mL tubes. When fixed samples are stored in 0.2mL PCR strip tubes or PCR plates, a styrofoam cooler can be used for slow cooling instead.
