Unique Dual Indices (UDI) are used during library preparation in Parse Biosciences workflows to uniquely label each sublibrary. This is achieved by applying a pair of barcodes, one on each end of the cDNA fragment, which helps ensure accurate sublibrary identification during sequencing.
UDIs play a crucial role in minimizing index hopping, a phenomenon that can occur during Illumina sequencing and result in the misassignment of reads to incorrect indices. By using UDIs, Parse workflows maintain high data integrity and reduce the risk of cross-contamination between samples.
Available UDI Plate Options
Parse Biosciences offers two UDI plate formats tailored to specific library types.

UDI Plate – WT (PN: UDI1001)
These libraries contain 5′ and 3′ TruSeq adapters, and the UDI Plate – WT adds TruSeq Read 1 and Read 2 sequences. -
UDI Plate – EC (PN: UDI1002)
These libraries contain 5′ Nextera and 3′ TruSeq adapters, and the UDI Plate – EC adds Nextera Read 1 and TruSeq Read 2 sequences.
UDIs serve as the 4th barcode during the Index PCR step. Please see the table below for more information on which plate(s) are required for each Parse® reagent kit:

UDI Plate Format and Compatibility
UDI plates are compatible with all Parse kit sizes and contain 48 single-use wells, each with a distinct UDI. Each sublibrary requires a unique UDI well.
As an example, for Evercode WT Kits, a single UDI plate can support up to:
- 24 Evercode WT Mini kits (2 sublibraries per kit)
- 6 Evercode WT kits (8 sublibraries per kit)
- 3 Evercode WT Mega kits (16 sublibraries per kit)
- 1.5 Evercode WT Penta kits (32 sublibraries per kit)
